Assou-Ekotto, 30, signed for Spurs from Lens in 2006, making 204 appearances and scoring four times for the White Hart Lane club before joining Queens Park Rangers on loan for the 2013-14 season.
The wing-back played 31 times for QPR as he helped Harry Redknapp's side achieve promotion to the Premier League, but has not played a single league match for Mauricio Pochettino's side this season.
A statement on the club's official website read: "We can confirm that Benoit Assou-Ekotto has been released from his contract with the club. We wish Benoit well for the future."
Assou-Ekotto featured for Cameroon at last summer's World Cup, though his solitary appearance saw him head-butt teammate Benjamin Moukandjo during the 4-0 group game defeat to Croatia.
